New findings about the prion protein and its interaction with the immune system

ScienceDaily (Dec. 29, 2011) ? Scrapie is a neurodegenerative disease which can function as a model for other diseases caused by an accumulation of proteins resulting in tissue malformations (proteinpathies), such as Alzheimer's and Parkinson's disease. Many questions regarding these diseases still remain unanswered. A new doctoral study has uncovered a number of factors relating to the uptake of the prion protein (PrPSc) associated with the development of this disease and how this protein interacts with the immune cells in the intestines.

Scrapie in sheep belongs to a group of diseases called "Transmissible spongiform encephalopathies"(TSE) because they are transmitted between individual animals and produce sponge-like, degenerative changes in the brain. These diseases afflict not only sheep but also cattle (BSE), deer (CWD) and humans (CJD). They can to a certain extent also be transmitted between species, as was the case during the 1990s, when over 200 people were infected by food and contracted CJD.

TSE, otherwise known as prion diseases, are thought to be transmitted by means of a diseased variant of a protein, the prion protein, which is a normal component of body cells and is most prolific in the brain. Generally speaking, prion diseases may be infectious, hereditary or occur sporadically/spontaneously. Disease arises when the normal prion protein mutates to the diseased variant, which differs from the healthy prion proteins by its change in structure. The body's cells have difficulty in breaking down this prion protein due to its different structure, and it therefore accumulates.

SInce PrPSc is to be found in the lymphatic tissue of the intestinal system at an early stage of the disease, it is assumed that transmission occurs via the gastrointestinal tract. During her doctoral research, veterinary scientist Caroline Piercey ?kesson studied the uptake of the prion protein in the intestines, thereby throwing new light on processes occurring during the early phase of the disease's development. Contrary to earlier assumptions, she demonstrated by means of immunoelectron microscopy that the prion protein responsible for the disease is not transported directly from the intestines to lymphatic tissue associated with the intestines. On the contrary, she showed that the protein passed freely or in lymphatic cells outside the organised lymphatic tissue in the intestines.

Dendritic cells are presumed to function as "gatekeepers" which determine what the body can tolerate and which immune defence reactions it needs to instigate when confronted with foreign substances. One of the objectives of ?kesson's project was therefore to examine the interaction of dendritic cells with prion protein uptake. Firstly, it was necessary to characterise dendritic cells in healthy sheep intestines and secondly, to investigate which types of cells were associated with the uptake of the prion protein.

Her findings showed that it was not dendritic cells, but macrophages, which were mainly responsible for the uptake of the protein. ?kesson's study revealed that the prion protein makes use of the normal physiological uptake channel for macromolecules in the intestines and that this may have a significant effect on the body's immunological surveillance system. One possible consequence is that immuno-tolerance is stimulated, thus impeding a normal immuno reaction against the prion protein absorbed via the intestines.

Future studies which can reveal how immunological cells are transported and how the prion protein is processed in the body will be of great interest, not only in order to provide more knowledge about scrapie, but also about other neurodegenerative proteinpathies, both in humans and animals.

Cand.med.vet. Caroline Piercey ?kesson defended her doctoral thesis on 20th December 2011 at The Norwegian School of Veterinary Science. The thesis is entitled: Studies on the uptake of prions and their early interaction with immune cells of the sheep gut.

GadgetTrak launches CameraTrace: Photo thieves beware (Digital Trends)

camertracerYou may remember hearing about photographer John Heller?s ordeal recovering his $9,000 worth of camera gear this summer. The thieves unwittingly uploaded photos taken with the stolen equipment, including a Nikon D3, to sites popular photo sharing platforms?which is just what GadgetTrak wanted.

The site was then testing a beta application that would scan photo-sharing sites like Flickr and 500px for images uploaded from potentially stolen equipment. The meta-data on these images reveals camera serial numbers, so owners missing their gear can easily identify if it?s theirs or not.

It was a dream come true for Heller, and now GadgetTrak is releasing the technology to consumers. Called CameraTrace, the service basically creates an identity for your camera so that in the cause it?s taken it can easily search the worldwide Web for any image taken with the device. Users pay a one-time $10 fee per camera, which comes with a lost-and-found tag that good Samaritans can use to trace back to owners who have merely misplaced their cameras.

In the case your device has been stolen, however, you will use the system to file a police report. GadgetTrak will even take up your case and speak with local enforcement if you choose. Basically you?ll have a helping hand through the painstaking process of getting your equipment back.

According to GadgetTrak, the CPUsage platform behind CameraTrace is able to thoroughly asses where your device has gone via uploaded images. It?s also allegedly able to identify more than 300 different ?high-end camera models? from various brand names.

Serious photographers will get an added benefit from using CameraTrace. The site also protects your copyrighted photos via its image monitoring service, which scans social sites to see if anyone out there is using your work without permissions?and pointing you in their direction.

Compared to how much photographers are willing to shell out on their gear, $10 is chump change. And given the popularity of the photo-sharing sites, this could be a very effective form of insurance.?

The poor diet of shift workers should be considered a new occupational health hazard, according to an editorial published in this month's PLoS Medicine. The editorial draws on previous work published in the journal, which showed an association between an increased risk of type 2 diabetes and rotating patterns of shift work in US nurses.

Shift work is now a very common pattern of work in both the developed and developing world, with around 15-20% of the working population in Europe and the US engaged in shift work. It is particularly prevalent in the health care industry. Shift work is notoriously associated with poor patterns of eating, which is exacerbated by easier access to junk food compared with more healthy options.

The editors argue that working patterns should now be considered a specific risk factor for obesity and type 2 diabetes, which are currently at epidemic proportions in the developed world and likely to become so soon in the less-developed world. They go on to suggest that firm action is needed to address this epidemic, i.e. that "governments need to legislate to improve the habits of consumers and take specific steps to ensure that it is easier and cheaper to eat healthily than not". More specifically, they suggest that unhealthy eating could legitimately be considered a new form of occupational hazard and that workplaces, specifically those who employ shift workers, should lead the way in eliminating this hazard.

Why Spotify Royalties Are Bigger Than Radio Royalties
-- Because of its large audience, radio is a more valuable revenue stream than on-demand subscription services. But when compared a revenue-per-listen basis, Spotify royalties exceed those paid by terrestrial radio in the U.S. and the U.K.? That's the conclusion of David Touve, an Assistant Professor of Business at Washington & Lee University who has long studied the music industry.

Per-listener, a spin on terrestrial radio ranges from $0.000186 to $0.000372 in the U.S. and from ?0.000299 to ?0.000498 in the U.K. ($0.0004 to $0.0007 at current currency rates), according to Touve's calculations. To arrive at these numbers Touve used publicly available financial and listening data. He incorporated many assumptions and needed to make one educated guess (the annual revenue of SESAC). His estimates cover a range of audience sizes for any given quarter hour as well as songs played per hours by radio stations. Note the U.K. figure includes both copyrights. In the U.S. only music publishers receive performance royalties for terrestrial radio.

Radio royalties may seem relatively large because people tend not to think of them in terms of revenue generated for transmitting a song to an individual listener. A single spin on terrestrial radio is heard by a large audience, and the performance royalties paid by a station is a function of its revenue. In contrast, payouts by on-demand services such as Spotify are reported as a per-play royalty. Thus, radio royalties seem big because many people are listening while on-demand royalties seem small because only one person is listening.

All four of Touve's radio estimates are far lower than what is typically paid out for a single play on Spotify (which splits a pool of revenue based on streaming activity). Take the range of U.S. terrestrial radio royalties of $0.000186 to $0.000372.? A typical Spotify payout of 0.3 cents per stream (an estimate based on Billboard sources and media reports) is 16.1 times greater than $0.000186 and 8.1 times greater than $0.000372. In addition, a 0.3-cent per stream royalty is nearly 3 times greater than the statutory royalty paid for a single stream by a non-interactive webcaster.

In other words, on-demand streaming generates more royalties for each time a single person hears a song. The biggest difference between radio and on-demand is the size of the audience. Radio royalties feel bigger because so many people listen to radio. And on-demand royalties feel small because so few people use services like Spotify.

An astute reader may note that a 0.3-cent Spotify payout is typical for the sound recording while U.S. terrestrial radio royalties cover only the composition. So this is a bit like comparing apples to oranges. But one should not easily dismiss the disparity between the estimates for radio and streaming royalties. Even if one adjusted radio royalties to include compensation for the sound recording, they would still lag behind streaming royalties. Indeed, Touve's estimates for U.K. radio royalties include both copyrights and still fall well behind streaming royalties.

Poor perception of streaming services' royalties can be explained, in part, by streaming services' paucity of listeners. As Touve notes, comparing royalties on a per-listen basis can change the perception of on-demand service's royaltyies. "Were the pool of money flowing from music services in the UK, such as Spotify, as large as that flowing from Radio, we might not be having these sorts of discussions," he writes. "Maybe someday both of these pools will be large. The bigger question than these present rates, therefore, is whether we should and how we might get to that version of someday."

BCS Bowl Games: All-SEC Championship An Embarrassment For College Football

The Alabama and LSU matchup in the BCS Championship Game this season is an embarrassment to college football and further proves that the sport badly needs a playoff system.

These two teams already played each other, and although it was a close, competitive game that either team could have won, it is extremely unfair that a team such as Oklahoma State will not have the opportunity to prove they are a title worthy team.

Stanford and Oregon are also teams who would certainly have a chance to beat LSU and Alabama. The Ducks could have beaten the Tigers in the season's opening week if not for some costly turnovers and sloppy play.

The Crimson Tide and Tigers may be the two best teams in the country, but how are we supposed to know if they really are the two best?

The Cowboys defense hasn't played great this season, but their offense is insanely talented and would have no problem putting up points on either of the Alabama or LSU defenses. In one game with everything on the line, the Cowboys defense could turn in an epic performance and shock one of the SEC powers with a strong defensive effort.

The big stage brings out the best in many players, and there are too many unknowns in the BCS.

We'll never know how good the Cowboys really are. We'll never know if Andrew Luck and his very good Stanford team could beat Alabama or LSU, and we'll never know if Oregon could overcome their struggles against SEC teams and beat LSU in a rematch.

An eight-team playoff system would give every deserving team a shot to prove they are the best team in the country.

It's foolish to punish the Cowboys for losing to an inspired Iowa State team in double overtime when they clearly deserve a shot at LSU.

Alabama had their shot to knock off Les Miles' undefeated Tigers and they failed. The fact that the Crimson Tide get another chance to beat LSU while several other teams don't get one chance is a travesty.

The BCS has proven time after time that it is too flawed a system to determine how the national championship of college football is won. This year's BCS title game matchup is just one example of the current system's failures, which cannot be tolerated any longer.

Pedestrian killed by car was 1st UNH female president

BEDFORD, N.H. -- Officials say a woman killed after she was struck while crossing a street was the first female president of the University of New Hampshire and Brandeis University.

Police say 78-year-old Evelyn Handler, of Bow, was killed Friday night while crossing a street in Bedford. She was found lying in the street and later died at a hospital.

UNH released a statement saying they were saddened to hear of Handler's death. They say she was president from 1980 to 1983.

Handler went on to become the first and only woman to lead Brandeis in Waltham, Mass., where she was president from 1983 to 1991.

No charges have been filed in Handler's death. Police are still investigating.

The fine folks at Verizon's Innovation Labs in Waltham, Massachusetts have posted a couple of videos of their efforts that include their vision of how Verizon plans to deliver TV to all the devices in your home, over both wired and wireless networks in HD, served off of a centralized media server. In addition to tablets, game consoles and computers, the media server will also serve content to smaller, more energy efficient thin clients at the other HDTVs in your home. Besides the obvious benefits of more content in more places, a centralized approach like this also offers the promise of being more user friendly by making the experience consistent across every screen -- no more walking to another room to schedule recordings. We're told the media server is expected to roll out late next year, but sadly, FiOS TV isn't exactly known for delivering new technologies when promised.

Amazon To Invest $270 Million In New Distribution Centers In Virginia, Tennessee

amazonOnline retail giant Amazon this morning announced that it will open four new distribution centers, two in Virginia and two in Tennessee, and that it will invest a total of $270 million to get the new facilities completed by next fall - just in time for next year's holiday season. In Virginia, Amazon says it will create about 1,350 jobs, while the new distribution centers in Tennessee will create 1,300 jobs. Amazon operates such facilities, which it calls fulfillment centers, to warehouse and ship items customers have ordered on Amazon.com.

Readiness for health, bioterror emergencies eroding: report (Reuters)

WASHINGTON (Reuters) ? Readiness for bioterror and health emergencies could erode nationwide as cash-strapped governments face cuts in spending, a study reported on Tuesday.

Key programs that detect and respond to bioterrorism, new disease outbreaks and disasters are at risk because of federal and state budget cuts, according to the ninth annual report on health preparedness by the non-partisan Trust for America's Health (TFAH) and the Robert Wood Johnson Foundation.

Among the programs, 51 of the 72 cities in the Cities Readiness Initiative, which supports distribution and administration of vaccines and medicine during emergencies, could be dropped.

The 10 state labs with top-level chemical testing status could lose that designation, the report said. The downgrade would leave the U.S. Centers for Disease Control and Prevention (CDC) as the only public health lab with full ability to test for chemical terrorism and accidents.

Twenty-four states are at risk of losing the support of career epidemiology field officers, who are CDC experts on responding to disease outbreaks and disasters.

The CDC's ability to mount an overall response to nuclear, radiologic and chemical threats as well as natural disasters is at risk because of potential budget cuts. All 50 states and the District of Columbia would lose the support CDC provides during these emergencies, the report said.

"We're seeing a decade's worth of progress eroding in front of our eyes," Jeff Levi, the Executive Director of TFAH, said in a statement.

Combined federal, state and local budget cuts mean public health departments can no longer support basic elements of preparedness, the report said.

The report in part urged assuring funding for public health readiness and improvement of biosurveillance. It also called for improved research, development and manufacturing of vaccines and medicines and improving the ability to care for an influx of patients in an emergency.

Michael Jackson house contents sell for nearly $1M

BEVERLY HILLS, Calif. (AP) ? The contents of the home where Michael Jackson lived with his three children just before his death have sold for nearly $1 million at auction.

Darren Julien, president of Julien's Auctions, was unable to provide a more specific figure as he continued to tally the totals Saturday after the daylong auction, which brought in nearly triple the company's pre-auction estimate of $200,000 to $400,000.

Among the highlights: A kitchen chalkboard where Jackson's children wrote "I love daddy," which sold for $5,000, and an armoire upon which Jackson wrote a message to himself on the mirror that fetched $25,750.

The auction also included furniture, artwork and other items from the rented mansion at 100 North Carolwood Drive, where Jackson lived as he prepared for a series of comeback concerts in London before his death in June, 2009.

The headboard from the bed where Jackson died at age 50 was removed from the auction at the family's request, but the rug that was beneath the bed sold for $15,360. The estimate had been $400 to $600.

Julien's Auctions re-created the mansion inside its Beverly Hills showroom and invited fans to fill the space where the bed would have been with a tribute to the late King of Pop. Julien promised to deliver all of the tribute items to Jackson's children and family matriarch Katherine Jackson.

"Michael Jackson has the greatest fans in the world. I can see why he lived for them," Julien said. "They came out every day this week to bring gifts. It's unlike anything we've ever seen as it relates to a celebrity and their fans."

Julien's Auctions has conducted auctions for dozens of celebrities, including Cher, Barbra Streisand, William Shatner and Slash.

Jackson commissioned the company to sell the contents of his Neverland Ranch before the auction was called off in early 2009. Julien's also sold Jackson's famous "Thriller" jacket for $1.8 million earlier this year.

Keystone pipeline looms large in U.S. tax-cut talks (Reuters)

WASHINGTON (Reuters) ? Lawmakers intensified efforts on Friday to extend payroll tax relief for Americans, and it appeared likely that language to speed approval of a controversial oil pipeline might be part of a deal.

With Senate leaders huddling in offices near the Senate chamber in search of a payroll tax cut deal, the House of Representatives separately approved a nearly $1 trillion funding bill to avert a government shutdown this weekend and keep federal agencies operating through to September 2012.

About one-third of House Republicans bucked Speaker John Boehner, however, voting against the bill because of concerns over its price tag.

The Democratic-controlled Senate was expected to approve the 1,209-page government funding bill on Saturday and quickly dispatch it to the White House for President Barack Obama to sign into law.

That would leave the payroll tax cut bill as the last major piece of business in a congressional session that has been dominated by acrimonious fights over taxes and spending. The partisan rancour has soured Americans on Congress.

Republicans and Democrats are now squabbling over how to pay

the $120 billion cost of the payroll tax cut. Republicans are standing firm on their demand that the Keystone XL pipeline be part of any deal, arguing it will be a major job creator.

Republican leaders want to expedite construction of the pipeline from Canada to Gulf of Mexico refineries in Texas. President Barack Obama has threatened to veto any coupling of the pipeline approval process to the payroll tax bill. But on Friday, the administration softened its tone.

White House spokesman Jay Carney, asked whether Obama would accept a bill that included a provision on the pipeline, said the most important thing was to get a bill done.

Obama has delayed a decision on the Keystone XL pipeline until 2013 and called for consideration of other routes to avoid ecologically sensitive areas. Environmental groups, a core Democratic constituency, strongly oppose the project.

But labor unions and some Democrats support it.

"Here's an opportunity for the president to say he's not going to let a few radical environmentalists stand in the way of a project that would create thousands of jobs and make America more secure at the same time," said Senate Republican leader Mitch McConnell.

TAX BREAKS, WAR SAVINGS

A Senate Democratic aide said negotiators were discussing ways to resolve the Keystone XL Pipeline issue, as well as how to make up for lost revenues if the payroll tax cut is continued for another year.

This could include killing a tax break on corporate jets and using savings from the completion of a drawdown of U.S. troops from Iraq by the end of the year, the aide said.

If Keystone becomes part of a deal, Obama and fellow Democrats will likely face the ire of environmentalists for abandoning their previous hardline stance. Earlier this week, Demorats also abandoned demands for a new surtax on the rich to pay for their payroll tax cut.

The payroll tax cut, first enacted one year ago, is set to expire on December 31. Democrats have been pushing for a one-year extension that would put about $1,000 more in the pockets of 160 million wage earners.

The White House and many economists argue that the payroll tax cut will help stimulate the struggling economy. Republican lawmakers question that but have grudgingly agreed to extend it.

Republican Representative Mike Simpson predicted the fight would soon be resolved.

"Both sides want to extend the tax cut, both sides want to extend the unemployment insurance. ... We all know those things have to happen, so we will come to a compromise."

Friday's negotiations were also focused on extending long-term unemployment benefits for millions of Americans and preventing a pay cut for doctors treating patients under the Medicare healthcare program for the elderly.
